Movie 23
Time-lapse confocal microscopy of mCherry-tubulin in tbg-1 (RNAi) in the one-cell stage of C. elegans embryos. Time-lapse movies were acquired using FV3000 Confocal system, 60X 1.4 NA objective, with high-sensitivity cooled GaAsP detection unit (Olympus Corporation, Japan) at 1 frame every 10 seconds with z-step size of 1 μm covering a thickness of 6 μm. Maximum z-projection is shown, played at 20 frames/sec. Note the drastic loss of the mCherry-tubulin fluorescence intensity at the centrosomes in this movie in comparison with control embryo (Movie 3). Also, note collapsing of the centrosomes after NEBD (compare with control embryo (Movie 3)).
Centrosome Aurora A regulates RhoGEF ECT-2 localisation and ensures a single PAR-2 polarity axis in C. elegans embryos
