Time-lapse confocal microscopy of mCherry-tubulin in air-1 (RNAi) embryo treated with Nocodazole in the one-cell stage of C. elegans embryos. Time-lapse movies were acquired using FV3000 Confocal system, 60X 1.4 NA objective, with high-sensitivity cooled GaAsP detection unit (Olympus Corporation, Japan) at 1 frame every 10 seconds with z-step size of 1 μm covering a thickness of 6 μm. Maximum z-projection is shown, played at 20 frames/sec. Note the drastic loss of the mCherry-tubulin fluorescence intensity at the centrosomes in this movie in comparison with control embryo (Movie 3). Also, note the failure of pronuclear meeting in air-1 (RNAi) embryos treated with Nocodazole (compare with Movie 3 (control) and S4 (air-1 (RNAi)).
