Time-lapse confocal microscopy of mCherry-PAR-2 in air-1 (RNAi); ect-2 (RNAi) in the one-cell stage of C. elegans embryos. Time-lapse movies were acquired using FV3000 Confocal system, 60X 1.4 NA objective, with high-sensitivity cooled GaAsP detection unit (Olympus Corporation, Japan) at 1 frame every 20 seconds, and played at 20 frames/sec. Note the loss of contractility in air-1 (RNAi); ect-2 (RNAi), as in ect-2 (RNAi) (Movie 35). Also note the delay in the rescue of single PAR-2 polarity axis. Compare with control embryo and air-1 (RNAi) embryo (Movies 33 and 34).
