Time-lapse confocal microscopy of mCherry-tubulin in air-1 (RNAi) in the one-cell stage of C. elegans embryos. Time-lapse movies were acquired using FV3000 Confocal system, 60X 1.4 NA objective, with high-sensitivity cooled GaAsP detection unit (Olympus Corporation, Japan) at 1 frame every 10 seconds with z-step size of 1 μm covering a thickness of 6 μm. Maximum z-projection is shown, played at 20 frames/sec. Note the drastic loss of the mCherry-tubulin fluorescence intensity at the centrosomes, and also collapsing of the centrosomes after NEBD in contrast to the control embryo (Movie 3).
