Time is indicated in hr:min. The peak of the pulse (EB stage) is set at 00:00. The movie starts 48 min prior to the peak of the pulse, but DAPT treatment had started already 12 h earlier and was continued during imaging. Since embryos share the same culture medium on the light-sheet microscope, the control embryo (Movie 5) was imaged in a different experiment.
