(related to Fig. 3H). Definitive macrophages are produced in the kidney niche of moonshine larvae by 6.5 dpf. The (gata2:gal4;UAS:Lifeact-GFP) transgene pair initially expressed by the aorta-derived HSPCs (somewhat mosaically) by 1.5-2.5 dpf still labels some of them and of their progeny, such as macrophages, labelled here by the mpeg1:mCherry-F transgene. The double-labelled cells are therefore definitive (HSPC-derived) macrophages - likely only a fraction of them that happen to still express GFP detectably. This is a z-scroll through a confocal z-stack (z-step=0.5 μm), lateral view, dorsal to the left, rostral to the top. The central panel shows the overlay of the green and red fluorescence channels, shown on either side, respectively. The proximal pronephric duct appears as a slightly autofluorescent epithelial structure through the last two thirds of the z-scroll.
