Live cell imaging of VAPB-downregulated NSC34 cells (clone #1).
Undifferentiated cells, expressing the PI4P-specific probe GFP-SidC, were incubated with Lysotracker, and then imaged in the green (PI4P) and red (Lysotracker) channels for ~6 min. While many doubly positive structures display little movement, rapidly moving vesicles positive for both components can also be observed. One example is highlighted by the white arrow in the 24.5 sec frame; two other examples are illustrated as a series of still images in Fig. 6B.
