DLD is compatible with long time-lapse imaging with minimal photobleaching and no apparent phototoxicity. CHO-K1 cells stably expressing paxillin-EGFP were seeded onto fibronectin-coated 35-mm glass bottom dishes and allowed to migrate for 16 hours in the presence of 0.0245 mW of incident light power. Images were captured with a Plan ApoChromat 20x/0.8NA objective and camera exposure time of 60,000 ms. Time is in hr:min. Scale bar is 25 μm.
