Videonanoscopy of JAMs for a JAM-Clow KG1 cell. Raw fluorescence (left) and trajectory reconstitution by MTT2col (right). MS5 cells were labeled with anti-JAM-B 829 and Alexa Fluor 488-conjugated anti-rabbit antibodies (green). KG1 cells were labeled with non-blocking rat 19H36 anti-JAM-C and Alexa Fluor 594-conjugated anti-rat antibodies (magenta). KG1 cells were dropped on MS5 cells before proceeding to video acquisition. Emission light was split on two Evolve 512 EMCCD cameras. 500 frames were grabbed with 100-ms exposure time. As indicated by timing, the movie is shown at live speed, 10 frames/s. Brightness and contrast were adjusted with FIJI to fit the range of intensities for each channel. Magenta trajectories were registered respectively to green trajectories (Fig. S2a). Trajectories are color-encoded according to their instantaneous diffusion, from dark (slow) to light (fast) green or magenta.
