Engineering Ply expression can govern Galectin8 dependent or independent killing. Time lapse imaging of SPN:Ply-High (S6 Movie) and SPN:Ply-Low (S7 Movie) SPN strains stained with DRAQ5 in YFP-Gal8 expressing A549s. Live cell imaging was performed under Laser scanning confocal microscope starting at 30 min p.i. S6 and S7 Movie depicts kinetics of Gal8 association with SPN:Ply-High and SPN:Ply-Low strains, respectively. Total 30 z-stacks were collected for both S6 and S7 movies, out of which MIP of stacks 10-30 (S6 movie) and 15-22 (S7 movie) were shown in the movies. Time interval, 15 min; scale bar, 5 μm.
