Representative single plane spinning disk microscopy time lapse showing collapse of lysosomal pH during LLOMe treatment. NRK cells were loaded with 100μg/mL of 10kDa FITC dextran and 100μg/mL 70kDa TMR dextran for 16 hours, then chased for 5 hours to lysosomes. Time lapse was captured for 15 minutes at 3 sec per frame. DMSO (left) or 1mM LLOMe (right) was added directly to stage at 3 minutes, indicated by black frames. Scale bar = 10μm, frame rate = 20 fps.
