Representative single plane spinning disk microscopy time lapse demonstrating pH collapse in DQ- late endosomes in response to LLOMe. NRK cells were loaded with 5μg/mL of DQ-BSA overnight, then loaded for 2 hours with 100μg/mL 10kDa Oregon Green dextran without chase. Time lapse was captured for 10 minutes at 3 sec per frame. DMSO (left) or 1mM LLOMe (right) was added directly to stage at 2 minutes, indicated by black frames. Scale bar = 5μm, frame rate = 20fps.
