Human single myofibre 3D segmentation in Imaris. The video displays the final Imaris segmentation and 3D model of a human gracilis single myofibre with an intact MTJ. The video frrst shows a full display followed by a Z-stack animation. The phalloidin staining and surface model (magenta), dystrophin staining (red), vimentin staining (green), nuclei staining (blue) are displayed. Spots of individual myonuclei (blue spheres) and “outside” nuclei of likely mononuclear cells (magenta spheres) are displayed. Myonuclei were defmed by both the localization within the dystrophin staining and lack of perinuclear vimentin staining. All other nuclei were labelled as “outside” nuclei. Scalebar 30 μm.
