Time-lapse confocal microscopy of ZYG-9-mCherry in air-1 (RNAi) in the one-cell stage of C. elegans embryos. Time-lapse movies were acquired using FV3000 Confocal system, 60X 1.4 NA objective, with high-sensitivity cooled GaAsP detection unit (Olympus Corporation, Japan) at 1 frame every 30 seconds with z-step size of 1 μm covering a thickness of 8 μm. Maximum z-projection is shown, played at 20 frames/sec. Note the drastic loss of the ZYG-9-mCherry fluorescence intensity at the centrosomes in air-1 (RNAi). Also, note the monopolar spindle formation. Compare with control embryo (Movie 41).
