Examples of the quantification approach for ectopic proliferation in fzd9b mutants in support of Figs 6B and 6E. Image stacks of new nephrons were rotated to be orthogonal to the plane of view. Then the lhx1a:eGFP+ new nephron was 3D cropped to retain only the 6 μm cell layer adjacent to the distal tubule. EdU+ nuclei were counted in this layer in wildtype, fzd9b mutants, and fzd9b mutants treated with the canonical Wnt inhibitor IWR1. The same new nephron as in movie 8 cropped to the bottom 6 μm volume revealing ectopic proliferation in basal layer cells.
